What to do for a week in Singapore

Although I have flown Singapore Airlines a few times I have only transited Changi without going into Singapore itself. Having Krisflyer points due to expire, my wife has always wanted to visit Singapore so we decided to use them for a Singapore trip flying Y on the way there and J on the way back with Singapore Airlines.

Our accommodation is booked at Clarke Quay so I am after any advice from those that know Singapore what they would recommend to see and do. I have googled this so have some ideas but would be interested in first hand experience.
