We're flying Melb-Singapore return with QF in Jun and then onto Sth Africa return with Emirates. We've bought 2 QF lounge passes with the original intention of using these before we fly back to Melb on QF. However going we have a 3 hour 15 min layover between the QF flight arriving Singapore and the Emirates flight leaving. Can you use lounge passes at the destination (Singapore) when we get off the QF flight?

I'm not familiar with rules about Lounge passes - but I would assume they are similar to eligibility into the QF Club as otherwise. Is your Emirates ticket sold as a QF flight number? If so I don't see why you can't use it.

The QF flights redeemed using mostly Qantas FF pts. The onward Emirates flights booked direct with Emirates so does not have a QF flight #. We would beh trying to enter the lounge in Singapore after flying the QF leg.

[Although the 2 airlines are partners you cannot upgrade an EK booking to the next class using QF pts for example]

You won't be able to use the passes in SIN prior to the EK flights.

There is no "arrivals" access with these passes.

So, no that plan will not work. You can link the passes to the QF flight ex-SIN though.
(it used to be that passes were not able to be used in SIN but this has changed)

Lounge Accessmeans access for Eligible Customers to an Eligible Lounge before the next onward Eligible Flight on that day.

Based on this, access is for the onward flight.

What class are you in for the EK flight Singapore to South Africa?

Though you might find that 3+ hours on the way there is shortened somewhat - maybe a late arrival in and you will need to be ready for boarding well before the departure time. Changi is quite pleasant to have a poke around - maybe the original plan to use them on the way home is the better one.
