I have an annual travel insurance policy with Colombus Direct, and paid my flights on the AMEX Platinum C/C (which has domestic travel insurance coverage).

Here's my dilemma; I'm flying up to Broome tomorrow morning to get some urgent work done, on that trip I also need to get up to Derby.

Cyclone Laurence has just been through and while downgraded, is dropping a reasonable amount of rain on the area. I've just rung Main Roads and the Great Northern Hwy is open, so I could be OK to get there and back. There is word the rain may worsen on the weekend though. If it does, and I get stuck in Derby is my insurance likely to cover me?

Not too sure what clause it may fall under in the policy.

You need to check with the insurance company. The Columbus Insurance, iirc, only covers you if you have pre-booked hotel iirc

If there is advice that the road may become impassable and you travel regardless, you may well find that you are on your on
