Officially, elite member beneifts do not apply for third party bookings. However it does not hurt to present your card at check-in, ask them to record it against your profile 'for incidentals points earn' and kindly ask for an upgrade. Expect nothing and anything offered is above and beyond what is required. I am a Platinum and have used this technique at three different properties in the past few years (Sheraton Waikiki, Sheraton Heathrow and Sheraton Wall Centre Vancouver) and received upgraded room and on site benefits eg. lounge acesss, but no points or stay credits.

Never hurts to ask. Don't forget that an "upgrade" (as in to a higher room category) isn't a Gold SPG benefit.

I've booked a few PL hotels and ended with Marriotts and Hilton. Hilton adds the HH number to booking but generally no points.

Marriott now reply

Please accept our apology, we are unable to add any information to Priceline.com reservations. We recommend adding your Marriott Rewards number when you check-in. Please note, reservations made through third party websites (such as Priceline.com), do not qualify for Marriott Rewards points or nights.

Have even had the Sheraton Anchorage give me the worst possible room in the house one night (priceline booking) then move me to a better room (direct booking) the next day.
Wasn't exactly impressed that they wouldn't combine.
YMMV

Funny that in one thread the 2 hotels that have given me the best upgrades are mentioned.when HH diamond put in the notes that it was mrsdrron's birthday(it was) and we got the Governor's suite(pre Sarah Palin).
The Westin Palace Madrid was the first hotel we stayed in as comped SPG plat.Upgraded to a 6 room suite overlooking the Square for the Spain Day parade.
But to the OP-definitely hand over your card but dont expect anything so you wont be disappointed. They do have a SPG Club Floor which would be all you could expect as Gold.

Oh I thought it did qualify for an upgrade based on this:
An enhanced room at check-in, when available.
But maybe im reading into it wrong

Just depends on how you define upgrade. Obviously some Hotels go above and beyond, but generally speaking it means within the same category so if you have room type x they may "enhance" it to room type x with a view over the local river/street/famous sight as opposed to the alley behind the hotel or give you room type x on the 22nd floor as opposed to room type x on the 2nd floor. But normally they won't upgrade you to room type z.

To be honest at the Palace I don't know what they view as "enhanced" rooms - I would have thought the street view as opposed to the courtyard view but apparently some people prefer the courtyard due to less noise etc.

The best rooms have a view over the Neptune fountain, you can also have rooms facing the main road up to the palace, the backstreet where the service vehicles come in or a laneway as well as the courtyard. The rooms are double glazed and our room was certainly very quiet when we closed the windows and doors. The club floor is on the top floor-the 6th I believe.
It's position is great with the Prado,Thyssen and Sofia Reina all within walking distance and the Hop on hop off bus starting just over the square.
