Just a quick question, trying to make sense of DONEX and DGLOBE fare rules, given their price is (slightly) less than DCIRC26 ex-AUS.

Is the following a valid routing? Australia-Asia-USA-Europe-Australia, with last part of journey either direct on QF2/6/10/30/32 or via JNB.

Forget JNB, that adds another Continent in Africa.

You can go directly from Europe via Asia in your routing as long as your stop in Asia is on a direct single flight number or a transit without stopover (<=24hours).
