I have booked a flight on the Qantas website to coughet next year. QF5557. It is a Jetstar flight. Is it possible to upgrade the flight with Qantas points or upgrade at all. As I booked with Qantas (paid). Qantas tells me to refer to the Jetstar web page.
 
No - from the FAQ:

What flights are eligible for Classic Upgrade Rewards?​

You can request an Upgrade Reward on eligible paid and confirmed tickets on flights operated by Qantas, with a Qantas (QF) flight number on the ticket.

QF5557 is what’s known as a codeshare flight. In other words QF sells the ticket (QF flight number) but not operate the flight (JQ operated).

So answer is no. But you can select better seats for a fee - see Jetstar seat selection

Upgrade at all... Yes, you can pay the full fare difference plus any change fees if applicable to upgrade to Jetstar Business. It's not going to be cheap, Qantas typically only sells full J fares on JQ codeshares which means the fare difference is probably going to be around ~$5032.

All that being said whilst Jetstar is still selling seats in J on their own website many of the QF codeshares in J are zeroed out (although plenty in Y fare types).

But at that point given J saver fares from MEL-SIN (QF J) then SIN-HKT (JQ Y) are about $3000 you'd be much better changing to that instead if trying to upgrade.
