Does anyone know if I can check my bags all the way through to Barcelona from Sydney. My first flight is on Singapore airlines through to Manila. Then I have a second separate ticket from Manila to Barcelona on Etihad Airways.

Just checked the SQ Checking in with partner airlines site, and it mentions:
Through check-in with partner airlines
With through check-in, you can check in for all your connecting flights to your final destination at the same time.
However, the Partner airlines list doesn't include EY.

Moreso, the above info on the SQ website would be pertaining to itineraries on a single ticket/PNR that includes travel on both SQ and a partner airline.

I would say that you'd have to re-check your bags at MNL, *however* you could ask nicely (plead) at the SQ check-in counter at SYD if they could kindly do it for you. Prepare for the worst though.

Source: Been through this kerfuffle before, with checking in at LHR at the LX counter, who weren't entertaining my request to be checked through on the itinerary LHR-ZRH-SIN-BLR, with LHR-ZRH on LX and ZRH-SIN-BLR on SQ, on separate tickets. Got it sorted at LHR eventually, but the amount of roadblocks (airblocks rather?) that we experienced throughout our time at the LHR airport has led us to never, ever try something like that again.
