Which airline to target ?

As my Citibank emirates card is now worthless I need to target another airline. Have a spend of 600,000 ++ a year and require 2x business class reward flights from Perth to UK per year from this . Which airline/credit card would give me the best chance ?

I'd be looking at a card that earns points transferrable to either Singapore Airlines KrisFlyer or Cathay Pacific Asia Miles as needed. Many of the major banks have these cards: ANZ, Westpac, HSBC, Amex branded cards, etc.

That way you have the choice of Singapore Airlines, Cathay Pacific, or any of their Star Alliance or oneworld partners. Redemptions are pretty good value with both programs, and the airlines are top quality too (and fly from Perth). Others may have other suggestions but that's what I would do.
