Can you squeeze one more little trip into your Hobart leg though?

Bruny Island - drive to Kettering for the ferry, about 10 k down the road make a quick stop at the Bruny Island Cheese factory for a tasting and supplies then on to Adventure Bay (south eastern side of Bruny) for the beach, then over to the South western side to the pub at Alonnah for the very best seafood platter (local oysters, salmon, smoked salmon, blue eye, calamari and whatever else is local and fresh).
(My son recently stayed nearby with the kids and went to the pub for tea - and was sent home with a complimentary mulled wine and two hot chocolates for the walk along the beach)

And if you do stay overnight somewhere, you can check out the penguins as they come ashore on 'the neck' of Bruny at dusk.

I think I already have Bruny Is and Adventure Bay marked on the map! I'll have to go and check!
 
OK, I think I've plotted everyone's suggestions onto Google maps and come up with a proposed route. An alternative in the North east would be Ansons Bay to Bridport (via Nth Ansons Rd) and then to George Town and the Tamar Valley.

Bridport is a nice seaside town, but I don't think there's a lot you'd get from going that way unless you want to play golf at Barnbougle (or see an aluminium smelter at Bell Bay). Pipers River Road where all the wineries links those two routes together, so you could do part of one and part of the other, rather than doubling back for 15km - it's not an actual routing, but you can see the relevant roads here
 
I know this is late to the thread. Heaps of great suggestions and i would not dispute any of them. A couple of options you might like to look at are the Tamar wetlands walk just outside Launceston. Its a real gem and only takes a couple of hours so can be done on the way back from some wine tasting. Also the Low Head penguin tours are as good as I've seen for getting up close to the little dudes in their natural environment.
