Would anyone know what Terminal SQ departs from at FCO and what time does check-in open for the midday flight to SIN.
 
All the international flights (apart from Schengen intra EU flights) check-in at the far end of the airport at check-in area C. It is usually fairly chaotic (hey, it's Rome!), and the contract check-in staff are fairly surly, though if you speak some Italian they sometimes brighten up.
I am pretty sure that all the trans-continental flights leave from the remote terminal for which you have to take the 'skytrain'. The announcements on the skytrain about doors closing, departing, arrival, and doors opening take almost as long as the ride!

Had previously looked at the web site but it doesn't go into detail as to which terminal SQ flies out of. However using a bit of lateral thinking, I did a search for the day's arrival info for the SQ flight and it shows Terminal C so I guess I've answered my own question.

My wife is leaving at 9.40am for Zurich & her UA award ticket itinerary says Terminal B. I guess we'll have to part ways at the Terminal B checkin and I'll have to use a shuttle to get to Terminal C check-in counters.

Check-in for all terminals is landside, you will only have to farewell your wife when you cross over to airside. The shuttle to the remote terminal is after you pass through security and outward immigration. In fact I think you could pass through security with your wife and then walk along the connecting corridor to terminal C where there is outward immigration.

That's good to know, thanks for the information.

The only catch will be is if check-in for my SQ flight opens early enough so that I can checkin and go thru with her as you suggest. Otherwise I'll have to hang around landside until the checkin counters open.
 
I haven't flown SQ from FCO (but have from elsewhere in Europe) but based on my experiences at FCO and other european airports, I would not expect check in to open more than 3 hours before the flight.
