If you are booked on a Qantas flight SYD-LHR as one flight number, I believe that meets the requirement for that tier.

As for a single sector, I don't believe one exists in OneWorld - not sure about other airlines, but can't think of any similar sectors either.

The highest mileage and closest single sector I could find is Singapore Airways' SIN-EWR (New York) (9535 miles non stop).
