Folks,

Back in Sept 2019, I booked Bris - LAX return flights for my wife and I, departing end of this month. In the meantime, my wife has had her Australian passport renewed,
It has recently arrived but has a new passport number.

Is this new passport number likely to cause any problems for us?
My wife is a worry wort and has delegated this question to me to follow up.
We will be taking a side trip to Canada and Mexico before catching the LAX to Brisbane return flight home

She will need a new ESTA for the US, as your ESTA is tied to the passport number. You should also update the passport details in My Booking (assuming a Qantas booking).
 
You'll need to update the new passport with the airline and get new ESTA and Canadian ETA.
US ESTA is valid for 2 years, or length of passport, whichever is shorter.
Canadian ETA is valid for 5 years, or length of passport, whichever is shorter.

If a passport has been replaced, for whatever reason, any attached visa (physical, electronic or waver) will, in most cases, need to be replaced. (There are some cases where a valid physical visa in an expired passport can be used, but that doesn't apply here)
 
For Aus Passport travellers, if travelling by air to or through Canada, you will need to obtain an online travel authorisation (eTA) before entering the country. As @Himeno pointed, needs new application with new Aus PP
