I had a search around and wasn't able to find anything on this.

Is it possible to book someone USA-AUS-USA? My wife's sister resides in the USA.

Or does the trip need to originate in AUS?

Many thanks.

Award flights can originate from anywhere, to anywhere*. They never need go near Australia!

*the qantas booking engine doesn’t allow travel to originate from a couple of cities around the world as they don’t have credit card facilities (or something like that). In those cases ticketing can be done via the Aussie call centre.
