I have a US trip coming up early May. At this stage the QF booking engine shows routing PER to SYD to DFW to RDU. This requires me to leave PER at either midnight or 5:45AM (with the required early rise)

What do you think the chance is of QF offering a routing PER-DXB-JFK to RDU?
 
Probably zero. I doubt that DXB-US is covered by the QF/EK partnership given QF's joint business with AA. If you were to book through EK you would find it, but would probably need JFK-RDU on a separate ticket. Wether or not you earn points and SCs on an EK booking remains very murky.

Going DFW to RDU is where I would go Expedia or equivalent as that would be pretty cheap in Aussie dollars.
The rest is pretty straight forward but we would avoid that midnight red eye that's for sure (out of Perth).
 
The timing of the flight from DFW to RDU actually works out really well, I did this last year and the connection worked out great. I wouldn't stress too much about the red-eye flight however, you have 15hrs to DFW to get that sleep...
 
I know that this is the QF forum, but I found AA have pulled back significantly from RDU, so you might want to consider at VA/DL combination as DL are now a significant operator to RDU and this was my best option from SYD last year. However I understand that this won't fit your preferred airline/ff program.
 
I'm sure if you do a multi-city booking on Qantas.com you can choose to leave Perth at a civilised time the day before your flight to DFW so you avoid the red-eye or 0545 departure. Of course this means you are up for a night of accommodation in Sydney but it might well be preferable top the alternatives!
