Woolworths allows you to withdraw money at their self check outs. Would this be likely to count as meeting the minimum spend if you were to simply withdraw $250 without buying anything?

Withdrawing using EFTPOS would mean pressing the 'savings" button on the eftpos machine, while the $250 minimum spend on the debit card would have to be done by pressing the 'credit' button on the eftpos machine so the spend would be recorded as being done via the Mastercard debit . The $250 spend can be done over multiple transactions, just make sure it is in the first 30 days after account opening.
